Modulation of Radioresponse by an Isoquinoline Alkaloid Berberine in Cultured HeLa Cells Exposed to Different Doses of γ –Radiation

چکیده
Ionizing radiations are used regulary to treat various tumors since the discovery of X-rays by Wilhelm Conrad Röntgen in the year 1895 and approximately 40% of patients are cured by radiotherapy and 50% of the patients receive radiotherapy during their disease [1,2]. It is used to treat tumors or as an adjuvant or neoadjuvant therapy and in several instances it is the only modality to treat tumors [3]. It is also used as a palliative therapy if the treatment is impossible [4,5]. Despite the success of radiotherapy its full potential remains underutilized due to the presence of hypoxic fraction of cells in the tumor and also due to the fact that with the increase in the radiation dose the normal tissue radiotoxicity increases [6]. The normal tissue toxicity can be alleviated and the tumor response to radiotherapy may be increased by combining certain drugs with radiotherapy.

Isoquinoline Alkaloid Berberine Exerts its Antineoplastic Activity by Inducing Molecular DNA Damage in HeLa Cells: A Comet Assay Study
Molecular damage of DNA plays an important role in the cell killing and several anti-neoplastic agents exert their cytotoxic effects by inducing DNA damage in the cancer cells. DNA damaging effect of various concentrations of berberine chloride (BCL), an isoquinoline alkaloid was studied in HeLa cells by alkaline comet assay. The DNA damage has been expressed as olive tail moment (OTM). Incubat...
متن کاملBerberine protects against glutamate-induced oxidative stress and apoptosis in PC12 and N2a cells
Objective(s): Neurodegenerative diseases have been associated with glutamatergic dysfunction. Berberine, an isoquinoline alkaloid broadly present in different medicinal herbs, has been reported to have neuroprotective effect. In the present study, the effects of berberine against glutamate-induced oxidative damage and apoptosis were investigated. Materials and Methods: The cultured PC12 and N2a...
متن کاملTrack detection on the cells exposed to high LET heavy-ions by CR-39 plastic and terminal deoxynucleotidyl transferase (TdT)
Background: The fatal effect of ionizing radiation on cells depends on Linear Energy Transfer (LET) level. The distribution of ionizing radiation is sparse and homogeneous for low LET radiations such as X or γ, but it is dense and concentrated for high LET radiation such as heavy-ions radiation. Material and Methods: Chinese hamster ovary cells (CHO-K1) were exposed to 4 Gy Fe-ion 2000 keV/...
متن کاملCharacterization of vacuolar transport of the endogenous alkaloid berberine in Coptis japonica.
Alkaloids comprise one of the largest groups of plant secondary metabolites. Many of them exhibit strong biological activities, and, in most cases, they are accumulated in the central vacuole of alkaloid-producing plants after synthesis. However, the mechanisms involved in alkaloid transport across the tonoplast are only poorly understood. In this study, we analyzed the vacuolar transport mecha...
متن کاملThe Impact of Hyperthermia in Radiosensitivity of Human Cervical Carcinoma Cell Line HeLa
Introduction: The objective of this work was to investigate the therapeutic effect of a combination of hyperthermia at 43oC exposured for 20, 40, 60 min with 6MeV electron beams at different radiation doses (2, 4, 6 Gy) compared to radiation alone. Materials and Methods: In this in vitro study, HeLa cells were exposed to either: 6MeV electron beams at diffe...
